400\">Developing a drug for Alzheimer\u2019s disease is a fundamental goal of the USC Center for Personalized Brain Health. We pursue a dual approach to advancing Alzheimer\u2019s treatment among carriers of the <em>APOE <\/em>\u03b54 gene variant, which magnifies the risk for the disease.<br \/>\n<\/span><\/p>\n<p><span style=\"font-weight: 400\">For those earlier in life, we focus on repairing a mechanism that is disturbed in<em> APOE <\/em>\u03b54 carriers: the body\u2019s delivery and use of lipids such as fatty acids, believed to be vital to keeping the brain healthy.<\/span><\/p>\n<p>For those later in life, we focus on excessive brain inflammation. Neuroinflammation is known to be a major cause of the degeneration seen in Alzheimer\u2019s disease.<\/p>\n<p>The Center for Personalized Brain Health launched its Alzheimer\u2019s drug development pipeline with a compound that shows promise for achieving both of these objectives. The perplexing answer was that increasing intake did not provide benefits. This suggested that there was an underlying mechanism that needed to be addressed.<\/p>\n<p>Subsequent studies showed that <em>APOE <\/em><span style=\"font-weight: 400\">\u03b54 <\/span>carriers accumulate more compounds in their bodies that break down omega-3s, compared to non-<em>APOE <\/em><span style=\"font-weight: 400\">\u03b54 <\/span>carriers. The effect was even more pronounced in <em>APOE <\/em><span style=\"font-weight: 400\">\u03b54 <\/span>carriers who had Alzheimer\u2019s disease.<\/p>\n<p>From there, we focused on an enzyme called cytosolic phospholipase A2 (cPLA2). Lab results suggested that cPLA2 was responsible for breaking down omega-3s for energy, rather than allowing them to support normal brain functions, such as memory formation. Crucially, inhibiting this enzyme in lab models increased the levels of omega-3 fatty acids in the brain and enhanced cognitive function.<\/p>\n<p>Working with colleagues at the USC Michelson Center for Convergent Bioscience, our team searched for a molecule likely to be safe and effective in blocking the action of cPLA2. Using computer modeling to screen a database of more than 18 billion compounds, we found BRI-50054.
